How many different conditions can you test at once? 

CRISPR screens, drug discovery, mRNA, and even the classic plasmids—the list of molecules that can be transfected using Nucleofector® technology is extensive. 

With over 25 years of innovation, Nucleofection technology is a powerful tool for high-throughput screening. This non-viral transfection method uses precise electric pulses to facilitate the rapid entry of substrates into both the cytoplasm and nucleus. Even when working with cell types that don’t yet have a validated protocol, there is an easy way to screen for the best possible transfection conditions. Moreover, the nucleofection process is remarkably swift, taking only a few seconds to complete. 


Different Nucleofector models—X unit, Y unit, 96-well unit, and up to the high-throughput 384-well Nucleofector system—offer varying capabilities for the number of samples and simultaneous conditions, ranging from 16 to 384 samples. An exciting example of the powerful application of nucleofection is the research performed by Wang et al. (2021, Cell), where they conducted a genome-wide CRISPR screen to identify viral entry factors in hosts for SARS-CoV-2 and other coronaviruses. This research was conducted using the Nucleofector 96-well unit. 

To summarize, nucleofection is a powerful technique for projects where you need to transfect a large number of cells in conditions that you already know will work, or if you are conducting a high-throughput screen. The right number of cells and conditions depends on your experiment or project.
